| Effect of Gastric pH on Oral Absorption of Poorly Soluble Weak Bases: Risk Assessment and Mitigation |
A large number of new chemical entities (NCEs) are poorly soluble (BCS class 2 or 4) and weak bases. These drugs tend to exhibit steep pH-solubility profiles - high solubility in stomach (low pH) and low solubility in gastrointestinal tract (high pH). Gastric pH perturbations due to disease state, food, or drug-drug interactions (e.g. antacids, H2blockers and proton pump inhibitors) could significantly alter gastric dissolution/solubility and thus the oral absorption. In certain cases the pH-dependent absorption could have a meaningful clinical impact (bioavailability, efficacy, and safety) requiring early intervention to mitigate the “pH-effect”. This presentation will focus on
• Factors influencing gastric pH and its impact on BA of weak bases.
• In silico, in vitro, and in vivo models to predict pH-dependent absorption.
• Formulation strategies to mitigate pH-effect. |
Balvinder Vig, PhD, Principal Scientist, Bristol-Myers Squibb Company
Dr. Vig is a Principal Scientist in Drug Product Science and Technology Department at Bristol-Myers Squibb Company. He received his Bachelor and Ph.D degrees in Pharmaceutical Sciences from University of Bombay, India and University of Maryland, Baltimore, respectively. Dr. Vig then pursued a postdoctoral research fellowship in the laboratory of Dr. Gordon Amidon at the University of Michigan, Ann Arbor. As a Research Fellow, he utilized transporter- and enzyme-based prodrug strategies and drug delivery approaches to improve oral absorption, stability, and targeting of anticancer and antiviral drugs.
Dr. Vig joined Bristol-Myers Squibb in 2003, where he is involved in the preformulation and formulation activities for new chemical entities; development of in vitro, in silico and in vivo models to understand the effects of physiological and formulation variables on drug absorption; and application of biopharmaceutical and drug delivery approaches to understand and improve developability of drugs.
Dr. Vig has presented extensively at national and international meetings and has over 19 publications in peer reviewed journals and 27 abstracts. He is an active member on several American Association of Pharmaceutical Scientists committees and has professionally contributed to American Chemical Society and American Peptide Society.
